HIV-derived antibacterial shows promise against drug-resistant bacteria

June 19, 2013 ? A team of researchers at the University of Pittsburgh has developed antibacterial compounds, derived from the outer coating of HIV, that could be potential treatments for drug-resistant bacterial infections and appear to avoid generating resistance. These new agents are quite small, making them inexpensive and easy to manufacture.

The research was published in the June 2013 issue of the journal Antimicrobial Agents and Chemotherapy.

The first of many probable applications will likely be the chronic bacterial infections in the lungs of cystic fibrosis patients "that frequently develop resistance to all standard antibiotics, and are the leading cause of death in these patients," says senior author Ronald Montelaro.

The lead compound shows powerful antibacterial activity against clinical isolates of diverse pathogenic bacteria that are resistant to most antibiotics. These agents, called engineered cationic antimicrobial peptides (eCAPs) "may be applicable to treatment of other respiratory infections, topical infections, and systemic infections," says Montelaro.

The genesis of the new agent was basic research on HIV envelope protein structure and function, says Montelaro. As part of this research, "we identified highly conserved unique protein sequences that were predicted by computer modeling to assume structures characteristic of natural antibacterial peptides. Since antibacterial peptides specifically target and disrupt the integrity and function of bacterial membranes, we thought that these similar peptide sequences in the HIV envelope protein might contribute to toxicity and death in infected cells by altering cell membranes."

The team engineered the original HIV peptides for greater effectiveness and smaller size, the latter to reduce manufacturing expenses. The engineering involved modifying amino acid content (they contain just two different amino acids), peptide length, charge, and hydrophobicity. The current paper describes the third generation peptides. The lead agent contains just 12 amino acid residues.

"Another potential application is biodefense, where eCAPs may be used as a rapid postexposure aerosol treatment in individuals after exposure to aerosolized pathogens, where the goal of immediate treatment would be to rapidly reduce bacterial dose from a lethal to a nonlethal or subclinical level," says Montelaro.

Obama to renew calls for nuclear reductions

BERLIN (AP) ? President Barack Obama will renew his call Wednesday to reduce the world's nuclear stockpiles, including a proposed one-third reduction in U.S. and Russian arsenals, the White House said.

Obama will make his case during a speech at Berlin's iconic Brandenburg Gate. His address comes nearly 50 years after John F. Kennedy's famous Cold War speech in this once-divided city.

The president has previously called for reductions to the stockpiles and is not expected to outline a timeline for this renewed push. But by addressing the issue in a major foreign policy speech, Obama is signaling a desire to rekindle an issue that was a centerpiece of his early first-term national security agenda.

The president discussed non-proliferation with Russian President Vladimir Putin when they met Monday on the sidelines of the Group of 8 summit in Northern Ireland. During Obama's first term, the U.S. and Russia agreed to limit their stockpiles to 1,550 as part of the New START Treaty.

It's unclear whether Obama will seek further reductions through a new treaty or a non-binding agreement that would not require congressional ratification. Republicans on Capitol Hill are likely to resist any further cuts to the U.S. stockpiles.

The Arms Control Association, a Washington-based policy group, urged Obama not to wait for a formal treaty, arguing the U.S. and Russian leaders could move more quickly by taking parallel steps to reduce warheads.

"Today's address is a good start but it is only a beginning," said Daryl Kimball, the group's executive director. "In the months ahead, President Obama must sustain the nuclear risk reduction enterprise and overcome petty partisan politics to help address today's grave nuclear challenges."

Obama's calls for cooperation with Moscow come at a time of tension between the U.S. and Russia, which are supporting opposite sides in Syria's civil war. Russia also remains wary of U.S. missile defense plans in Europe, despite U.S. assurances that the shield is not aimed at Moscow.

Also in Wednesday's speech in Berlin, Obama will press Congress to pass a nuclear test ban treaty, the White House said, seeking to revive an effort that has stalled in recent years.

Germany's foreign minister, Guido Westerwelle, is a strong advocate of nuclear disarmament and has long called for the removal of the last U.S. nuclear weapons from German territory, a legacy of the Cold War. The Buechel Air Base in western Germany is one of a few remaining sites in Europe where they are based.

Under an agreement drawn up when they formed a coalition government in 2009, Merkel's conservatives and Westerwelle's Free Democratic Party agreed to press NATO and Washington for the nuclear weapons to be withdrawn, but did not set any timeframe.

Nuclear stockpile numbers are closely guarded secrets in most nations that possess them, but private nuclear policy experts say no countries other than the U.S. and Russia are thought to have more than 300. The Federation of American Scientists estimates that France has about 300, China about 240, Britain about 225, and Israel, India and Pakistan roughly 100 each.

Twitter Buys Boston-Based Spindle, Team Heading to S.F.

Twitter?s gone shopping in Boston again.

Spindle, a Boston-based startup that culls social-media messages to give consumers a view of what?s happening nearby, announced today that it has been acquired by Twitter.

Terms of the deal were not disclosed. It appears to be a ?talent acquisition,? since the Spindle service is shutting down and its team relocating to San Francisco.

Spindle had raised $2.3 million in seed funding?from a long list of investors:?Polaris Ventures, Greylock Partners, Lerer Ventures, SV Angel, Atlas Ventures, Broad Beach Ventures, and Project 11, along with former Microsoft technical chief Ray Ozzie and Raman Narayanan.

Twitter?s other two Boston-area acquisitions?advertising data service Bluefin Labs and mobile-app analytics startup Crashlytics?have both stayed in Boston, forming the basis of Twitter?s office here. We reported that Twitter spent more than $100 million (in cash and stock) for Crashlytics, and around $100 million for Bluefin Labs.

Spindle was founded by former Microsoft employees, including CEO Pat Kinsel. The startup was among the?software companies trying to perfect local search on mobile devices by tapping into the flood of information being generated by companies and everyday people.

At the base of this problem is a pretty basic behavior for the smartphone age: A consumer whipping out their sensor-laden mobile device, trying to figure out where to eat or what to do for fun when they?re out on the town.

Nobody?s really hit on the solution yet, although some big names have tried for years?Google, Foursquare, Yelp, Facebook, and quite a few startups. By buying Spindle, you can assume that Twitter is hoping to get more serious about local search, too.

?By joining forces with Twitter, we can do so much more to help you find interesting, timely, and useful information about what?s happening around you,? Spindle said in its blog post.

Earlier this year, CEO Kinsel told me that Spindle believed its technology was solid enough to create plenty more applications beyond aggregating social-media streams for mobile users. ?We really believe strongly in this app. But on the technology side, we?re trying to do much more,? Kinsel said.
